Retatrutide 30mg: A Triple-Agonist Research Guide
Retatrutide (LY3437943) is an investigational peptide that acts as an agonist at three receptors at once: the glucagon-like peptide-1 receptor (GLP-1R), the glucose-dependent insulinotropic polypeptide receptor (GIPR) and the glucagon receptor (GCGR) (Coskun et al., 2022). That single-molecule, three-receptor design makes it a useful tool compound for laboratories studying how incretin and glucagon signaling interact in energy balance, glucose regulation and lipid metabolism.
This guide summarizes the published receptor pharmacology and preclinical findings, explains how retatrutide differs from the GLP-1 receptor agonist semaglutide and the dual GIP/GLP-1 receptor agonist tirzepatide, and covers practical points for in vitro and animal-model work. BioSim’s retatrutide is supplied strictly for laboratory research. It is not a medicine, it is not for human or veterinary use, and nothing here is guidance on administration to people.
What Makes Retatrutide Unique?
Most incretin-based research compounds act on one receptor (semaglutide at GLP-1R) or two (tirzepatide at GIPR and GLP-1R). Retatrutide adds glucagon receptor agonism. In the discovery paper, retatrutide showed balanced GCGR and GLP-1R activity in vitro, with comparatively greater activity at GIPR (Coskun et al., 2022). That relative balance between the three receptors is the property researchers most often need to account for when interpreting results, because each receptor has different tissue distribution and downstream effects.
The Three Targets
- GLP-1 receptor: GLP-1 is an incretin hormone. Published reviews describe incretin receptor signaling as regulating islet hormone secretion, glucose concentrations, gut motility, appetite and body weight, with effects in many tissues through direct and indirect neural and endocrine pathways (Campbell & Drucker, 2013).
- GIP receptor: GIP is the second major incretin. The same review covers its roles in islet hormone secretion and lipid metabolism, and highlights unresolved questions in GIP biology that remain active research topics (Campbell & Drucker, 2013).
- Glucagon receptor: glucagon is best known for raising blood glucose, but recent work has drawn attention to its less appreciated effects on lipids and body weight, which is why glucagon agonism has been incorporated into multi-receptor research compounds (Müller et al., 2017).
Preclinical Findings
The key preclinical data come from the discovery study. In obese mice, retatrutide decreased body weight and improved glycemic control. The authors attributed the larger body-weight effect to the combination of two mechanisms: reduced calorie intake driven by GIPR and GLP-1R agonism, plus increased energy expenditure driven by GCGR agonism (Coskun et al., 2022).
For a laboratory, that finding frames the main experimental questions. How much of a measured effect comes from intake and how much from expenditure? Which receptor is responsible in a given tissue? Studies that pair retatrutide with selective GLP-1R or GIPR agonists, receptor antagonists, or receptor-knockout models can help separate these contributions. Retatrutide remains an investigational compound, and the discussion here is limited to receptor pharmacology and animal-model data.
“GLP-3”: A Note on Terminology
Retatrutide is sometimes called “GLP-3” online. This is an informal label rather than a scientific term. There is no GLP-3 receptor or hormone in the literature cited here; the discovery paper describes retatrutide as a triple agonist at the glucagon, GIP and GLP-1 receptors (Coskun et al., 2022). Using the receptor-based description in lab notes and publications avoids confusion.
Retatrutide vs. Semaglutide vs. Tirzepatide
Semaglutide and tirzepatide are the two reference compounds most often used for comparison. Understanding how they were designed helps clarify what retatrutide adds.
Semaglutide: a selective GLP-1 receptor agonist
Semaglutide was designed as a long-acting analog of human GLP-1. It has two amino acid substitutions compared with human GLP-1 (Aib8 and Arg34) and carries a fatty-acid derivative at lysine 26 that increases albumin binding. In mini-pigs, its plasma half-life after intravenous administration was 46.1 hours (Lau et al., 2015). Reversible albumin binding was the main strategy used to extend the action of both liraglutide and semaglutide (Knudsen & Lau, 2019).
In rodent studies, semaglutide reduced food intake and body weight without decreasing energy expenditure. Semaglutide reached the brainstem, septal nucleus and hypothalamus but did not cross the blood-brain barrier; it accessed the brain through the circumventricular organs and selected sites next to the ventricles, and it induced c-Fos activation in 10 brain areas (Gabery et al., 2020).
Tirzepatide: a dual GIP and GLP-1 receptor agonist
Tirzepatide (LY3298176) is a fatty-acid-modified peptide with dual GIP and GLP-1 receptor agonist activity. In mice, it activated both receptors’ signaling in vitro, showed glucose-dependent insulin secretion, and with chronic administration decreased body weight and food intake (Coskun et al., 2018). Later pharmacology work found that tirzepatide is imbalanced, engaging the GIP receptor more than the GLP-1 receptor, and biased at the GLP-1 receptor, favoring cAMP generation over β-arrestin recruitment (Willard et al., 2020).
Side-by-side receptor profile
| Compound | GLP-1R | GIPR | GCGR | Key published pharmacology |
|---|---|---|---|---|
| Semaglutide | Agonist | No | No | Acylated GLP-1 analog with albumin binding (Lau et al., 2015) |
| Tirzepatide | Agonist (biased) | Agonist (greater engagement) | No | Imbalanced toward GIPR; cAMP-biased at GLP-1R (Willard et al., 2020) |
| Retatrutide | Agonist | Agonist (greater activity) | Agonist | Balanced GCGR/GLP-1R activity in vitro (Coskun et al., 2022) |
The practical difference for researchers is glucagon receptor agonism. Semaglutide and tirzepatide studies in mice point mainly to reduced intake (Gabery et al., 2020; Coskun et al., 2018), while the retatrutide discovery study adds a GCGR-linked increase in energy expenditure (Coskun et al., 2022). Comparative designs that include all three compounds can isolate that extra mechanism.
